Microspectrophotometry












A microscopic thin beam of light enters the outersegment of the receptor. Some of the light is absorbed by the photopigment and the rest emerges to be captured by the very sensitive detector.

The difference between that which enters the outersegment and that which emerges is the amount that is absorbed.
